High temperatures, whether of shoot or root, are reported to affect shoot and root growth of various plant species. The scanty information available on the differential response of wheat genotypes to high shoot and root‐zone temperatures triggered this investigation to study the response and adaptation of shoot and root growth of three wheat genotypes to high shoot and root‐zone temperatures during early growth stages. Three wheat genotypes; Fang (heat tolerant), Siete Cerros (heat sensitive) and Imam (recent cultivar adapted to a hot irrigated environment) were grown in soil and hydroponically. Three shoot/root‐zone temperatures (23/23, 23/35 and 35/35 °C for the soil experiment and 22/22, 22/38 and 38/38 °C for the hydroponic experiment) were applied at three‐leaf growth stage. High root‐zone temperature alone or combined with high shoot temperature reduced xylem sap flow rate, root dry weight, root length and root/shoot ratio. Unexpectedly, shoot fresh and dry weights and relative growth rate (RGR) were not significantly affected by the high root‐zone temperature except for the susceptible genotype, Siete Cerros, after prolonged exposure in the hydroponic experiment. In contrast, high shoot/root‐zone temperature significantly reduced shoot fresh and dry weights from as early as the first week of the hydroponic experiment. The 38/38 °C treatment also caused significant reduction in RGR and net assimilation rate during the first 2 weeks, but no significant differences were found during the last 2 weeks compared with 22/22 °C. Interesting responses were observed among genotypes in terms of shoot and root dry weights and root/shoot ratio at 38/38 °C treatment. The heat‐sensitive Siete Cerros showed the least reduction in these traits during the first 2 weeks while the heat‐tolerant Fang and Imam responded by greatly reducing their shoot and root weights. The situation was almost reversed with the duration of treatments such that Siete Cerros became the most affected genotype while Fang and Imam were better adapted to high shoot/root‐zone temperature. Specific root weight was the exceptional trait that increased under high temperature treatments. Results indicate that despite the reduction in root length and weight observed under high root‐zone temperature, shoot growth was not much affected suggesting that the use of suitable cultivar coupled with proper management could alleviate most of high root‐zone temperature effects during early growth stages.  相似文献

 Nitrous oxide (N2O) emissions were measured from an irrigated sandy-clay loam cropped to maize and wheat, each receiving urea at 100 kg N ha–1. During the maize season (24 August–26 October), N2O emissions ranged between –0.94 and 1.53 g N ha–1 h–1 with peaks during different irrigation cycles (four) ranging between 0.08 and 1.53 g N ha–1 h–1. N2O sink activity during the maize season was recorded on 10 of the 29 sampling occasions and ranged between 0.18 and 0.94 g N ha–1 h–1. N2O emissions during the wheat season (22 November–20 April) varied between –0.85 and 3.27 g N ha–1 h–1, whereas peaks during different irrigation cycles (six) were in the range of 0.05–3.27 g N ha–1 h–1. N2O sink activity was recorded on 14 of the 41 samplings during the wheat season and ranged between 0.01 and 0.87 g N ha–1 h–1. Total N2O emissions were 0.16 and 0.49 kg N ha–1, whereas the total N2O sink activity was 0.04 and 0.06 kg N ha–1 during the maize and wheat seasons, respectively. N2O emissions under maize were significantly correlated with denitrification rate and soil NO3 -N but not with soil NH4 +-N or soil temperature. Under wheat, however, N2O emissions showed a strong correlation with soil NH4 +-N, soil NO3 -N and soil temperature but not with the denitrification rate. One of the basic steps in objective analysis of sperm motility is the subdivision of a motile sperm population into slow, medium and rapid categories based on their velocity. However, for CASA analysis of quail sperm, the velocity values for categorization of slow, medium and rapid sperm have not yet been standardized. To identify the cut‐off values of “velocity curvilinear” (VCL) for quail sperm categorization, we captured and analysed 22,300 tracks of quail sperm using SCA®‐CASA. The median and mean VCL values were 85 and 97 μm/s. To define the VCL cut‐off values, we used two methods. In the first, we identified the upper (rapid sperm) and lower (slow sperm) cut‐off values using: (i) median VCL ± 25% or ± 50% or ± 75% of median VCL value; (ii) first and third quartile values of VCL data (i.e. 25% cut‐off setting); and (iii) 33% and 66% of VCL data. Among these settings, sperm categories and their corresponding motility characteristics recorded using the “25%” setting (i.e. slow ≤36 ≤ medium ≤154 ≤ rapid) were found the most realistic and coherent with male ranking by fertility. In the second method, we calculated heteroscedasticity in the total VCL data using PCA and the two‐step clustering method. With this approach, the mean of the high and low clusters was 165 and 51 μm/s, respectively. Together, the mean from two methods suggested that, for SCA®‐CASA categorization of quail sperm, sperm should be classed as “rapid” at VCL ≥160 μm/s and “slow” at VCL ≤45 μm/s.  相似文献

Plant regeneration ability was studied in the medicinal-ornamental plant, Cosmos bipinnatus ‘Sonata white’, which is a dicotyledonous recalcitrant plant to shoot induction. Cotyledons were used as sources of explants to investigate plant regeneration. High frequency of direct shoot induction was obtained when BA (5 mg/l) and AgNO3 (5 mg/l) were used in combination with 20 mg/l adenine sulphate (73.8%) in Murashige and Skoog (MS) medium. The highest shoot number per explant (5.7) was induced on MS medium supplemented with 5 mg/l BA, 5 mg/l AgNO3, and 40 mg/l adenine. Eight week-old shoots were transferred to root induction media containing MS and half-strength MS medium with different concentration of IBA. The highest rate of root induction (70.8%) was obtained on half-strength MS medium with 1.5 mg/l IBA within four weeks. The plantlets were transferred to pot and kept in the greenhouse condition. Seventy percent of the plantlets successfully acclimatised.

The most significant focal points of the embryo transfer technology are as follows: the selection of donors, the response of the selected donor to the superovulation protocol and the obtained number of the transferable embryos. For this purpose, it is suggested that donor selection can be done by anti‐Müllerian hormone (AMH) levels, and embryo production is evaluated. AMH is secreted by the granulosa cells of primordial, pre‐antral and antral follicles below 4 mm in the ovary, independent of FSH. Therefore, the aim of this study was to investigate the relationship between serum AMH levels and the number of corpus luteum (CL), total embryos and transferable embryos that were shaped after a uniform superovulation protocol. For this reason, 48 Simmental cows, which were located at General Directory of Agricultural Enterprises (region, province, etc. instead of the general directorate), were used as donors for the embryo transfer. Blood samples were taken at random, regardless of the stage of animal's sexual cycle. AMH levels were measured by enzyme‐linked fluorescent assay (ELFA) method of the miniVIDAS® (bioMérieux SA) using AMH Bovine Test Kit. According to the statistical analyses of the obtained data, AMH levels were positively correlated with CL and total embryos (p < .05). No significant correlations between AMH and transferable embryos were approved (p > .05). It was also determined that each 200 pg/ml increase in serum AMH level resulted in one increase in CL number. Overall, considering the positive correlation between AMH level and the obtained number of CL and total embryos after a superovulation treatment, it was concluded that measuring blood AMH level prior to any further costly implementation may be an effective method in donor selection.  相似文献

Ornithogalum ulophyllum Hand.-Mazz. with beautiful white flowers is an important medicinal and ornamental plant of the Middle Eastern countries and need exploitation for commercial propagation. The study reports in vitro mass proliferation of bulblets achieved from twin scales and “in vitro regenerated bulblet” explants on MS medium supplemented with various concentrations of BAP–NAA. The best regeneration on twin scales and “in vitro regenerated bulblets” was obtained on MS medium containing 2 mg l−1 BAP–0.5 mg l−1 NAA and 2 mg l−1BAP–1 mg l−1 NAA, respectively. However, bulb scales seemed to be more potent for bulblet regeneration. A large number of the developing bulblets rooted on the regeneration medium. Remaining non-rooting bulblets were rooted on MS medium containing 1 mg l−1 NAA. All plants were acclimatized in the environmental chamber for 4 weeks and were transferred to the greenhouse for flowering. Regenerated bulblets developed into morphologically normal plants.  相似文献

Agar is being used to solidify media for plant tissue culture since long. Both purity and type of agar or gelling agent influence the behaviour and growth of tissues in culture. The study using leaf disc explants of variety Samsun of tobacco compares adventitous shoot regeneration on 0.5 cm × 0.5 cm leaf discs cultured on MSD4X2 medium and rooting on MSO medium gelled with different blends of agar–isubgol, gelrite–isubgol, phytagel–isubgol or isubgol singly. It was found that irrespective of some problems associated with isubgol gel, the maximum number of shoot per explant were recorded on MSD4X2 medium gelled with 7 g/l isubgol. The longest shoots were recorded on MSD4X2 medium gelled with 9 g/l isubgol. Likewise, the highest number of roots were also recorded on MSO medium gelled with 7 g/l isubgol. For a given quantity of a medium, agar, gelrite/isubgol blends are very cheap compared to agar used singly. Moreover, blends of agar/isubgol, gelrite/isubgol gelled at low temperatures indicated safe use of these gels for heat labile substances in genetic transformation or tissue culture experiments. The results emphasized the potential of the isubgol used singly or in combination with agar and gelrite for economic commercial application, replacing the costliest, though not indispensable, gelling agent agar.  相似文献

Crimean–Congo Haemorrhagic Fever (CCHF) is a deadly viral zoonotic disease, which is endemic in Pakistan. We report a case study analysis of three cases of CCHF which occurred in Chakwal, Pakistan in 2016. The disease was suspected in three patients exhibiting clinical symptoms suggestive of CCHF; two of the three patients died. The presence of CCHF was detected by seroconversion (IgG) from the surviving patient, while the antigen was detected in Hyalomma ticks originating from animals in the vicinity. This report indicates increase threat emergence of CCHF in Pakistan and highlights its zoonotic implications requiring immediate interventions under the “One‐Health” concept.  相似文献
